Diffuse Intrinsic Pontine Glioma, commonly referred to as pontine glioma, infiltrative brainstem glioma, or DIPG, is a rare tumor of the brainstem that occurs almost exclusively in children. A pontine glioma occurs in a most delicate area of the brainstem (the "pons"), which controls many critical functions, including breathing and blood pressure. The majority of pontine tumors are diffuse intrinsic brainstem gliomas, which are usually high grade, locally infiltrative, and have a uniformly poor prognosis [ 9 ]. Histologically, these tumors are usually anaplastic astrocytomas (World Health Organization [WHO] grade III) or glioblastoma (WHO grade IV). Diffuse intrinsic pontine glioma (DIPG) is a lethal pediatric brain tumor and the leading cause of brain tumor–related death in children. As several clinical trials over the past few decades have led to no significant improvements in outcome, the current standard of care remains fractionated focal radiation. Symptoms of Diffuse Intrinsic Pontine Glioma typically present for approximately 1 month before diagnosis, but subtle symptoms may be present for up to 6 months prior to diagnosis. Because of their location in the brainstem, as they grow, DIPGs cause pressure on the cranial nerves that originate in the pons.

The pons controls essential bodily functions such as heartbeat, breathing, swallowing, eye movement, eyesight, and balance. H3K27M-mutant gliomas, including diffuse intrinsic pontine gliomas (DIPGs), are lethal childhood brain tumors. DIPGs are inoperable due to their location within the pons/brainstem. Available treatment options, including chemo-/radiotherapy, are ineffective, and over 90% of patients die within 1.5 years of diagnosis (Morales La Madrid et al., 2015 2017-05-08 · Diffuse intrinsic pontine glioma (DIPG) is a universally fatal pediatric cancer.

MRI shows infiltrative expansion of the pons that is typically hypointense on T1-weighted images, hyperintense on T2-weighted and FLAIR images, and has no significant dorsal exophytic component. Tribute to all the boys who battled DIPG and have now sadly passed away. This type of cancer is so rare that these are the only boys that I am aware of who h 2020-11-10 · Background Diffuse intrinsic pontine glioma (DIPG) is the most common brainstem cancer in childhood. This rapidly progressing brainstem glioma holds a very dismal prognosis with median survival of less than 1 year.
Radiation is the only effective treatment and extends overall survival to a median of 11 months. Due to its location in the brainstem, DIPG cannot be surgically resected. Diffuse intrinsic pontine glioma (DIPG) is a brain tumor found in a part of the brain stem called the pons.

The majority of cases of DIPG exhibit a mutation in histone-3 (H3K27M) that results in oncogenic transcriptional aberrancies. We show here that DIPG is vulnerable to transcriptional disruption using bromodomain inhibition or CDK7 blockade. Diffuse intrinsic pontine glioma (DIPG), a high-grade glioma that arises in the pons is predominantly seen in children. This central nervous system (CNS) malignancy represents the leading cause of brain-tumor related death . Ionizing radiation extends overall survival to a median of 11 months .

Diffuse Intrinsic Pontine Glioma (DIPG) Diffuse intrinsic pontine gliomas (DIPG) are highly aggressive and difficult to treat brain tumors found at the base of the brain. They are glial tumors, meaning they arise from the brain's glial tissue — tissue made up of cells that help support and protect the brain's neurons.
